Autophagy is the major pathway for the delivery of cytoplasmic material to the vacuole or lysosome. Selective autophagy is mediated by cargo receptors, which link the cargo to the scaffold protein Atg11 and to Atg8 family proteins on the forming autophagosomal membrane. We show that the essential kinase Hrr25 activates the cargo receptor Atg19 by phosphorylation, which is required to link cargo to the Atg11 scaffold, allowing selective autophagy to proceed. We also find that the Atg34 cargo receptor is regulated in a similar manner, suggesting a conserved mechanism.  相似文献

Lytic polysaccharide monooxygenases (LPMOs) are a recently discovered class of enzymes that employ a copper-mediated, oxidative mechanism to cleave glycosidic bonds. The LPMO catalytic mechanism likely requires that molecular oxygen first binds to Cu(I), but the oxidation state in many reported LPMO structures is ambiguous, and the changes in the LPMO active site required to accommodate both oxidation states of copper have not been fully elucidated. Here, a diffraction data collection strategy minimizing the deposited x-ray dose was used to solve the crystal structure of a chitin-specific LPMO from Enterococcus faecalis (EfaCBM33A) in the Cu(II)-bound form. Subsequently, the crystalline protein was photoreduced in the x-ray beam, which revealed structural changes associated with the conversion from the initial Cu(II)-oxidized form with two coordinated water molecules, which adopts a trigonal bipyramidal geometry, to a reduced Cu(I) form in a T-shaped geometry with no coordinated water molecules. A comprehensive survey of Cu(II) and Cu(I) structures in the Cambridge Structural Database unambiguously shows that the geometries observed in the least and most reduced structures reflect binding of Cu(II) and Cu(I), respectively. Quantum mechanical calculations of the oxidized and reduced active sites reveal little change in the electronic structure of the active site measured by the active site partial charges. Together with a previous theoretical investigation of a fungal LPMO, this suggests significant functional plasticity in LPMO active sites. Overall, this study provides molecular snapshots along the reduction process to activate the LPMO catalytic machinery and provides a general method for solving LPMO structures in both copper oxidation states.  相似文献

The cytochalasin B binding component of the human erythrocyte monosaccharide transport system has been purified. The preparation appears to contain one major protein with an apparent polypeptide chain molecular weight of 55 000 and about 0.4 binding sites per chain. Cytochalasin B binds to the reconstituted preparation with a dissociation constant of 1.3·10?7 M, a value which is similar to that reported for the transport system in the intact erythrocyte.  相似文献

The notion that the individual and society articulate only tenuously, if not with tension, may be social scientific folklore. Human aptitudes involving creativity, independent thought, and agency generally, appear to be shaped and sometimes dictated by the socioculture. Social scientists have begun to unpack the issue of agency in order to better understand the relations among and between individuals and society. By exploring an empirical case involving a congregation of Svenska Missionsförbundet, one can establish that while the form of religious practice is individualistic, its content is strikingly communalistic. These practitioners, and other Swedes at large, often perceive a contradiction between the agendas of individual and of community, which conflict bears critical scrutiny.  相似文献

While yoga is gaining increased popularity in North America and Europe, its safety has been questioned in the lay press. The aim of this systematic review was to assess published case reports and case series on adverse events associated with yoga. Medline/Pubmed, Scopus, CAMBase, IndMed and the Cases Database were screened through February 2013; and 35 case reports and 2 case series reporting a total of 76 cases were included. Ten cases had medical preconditions, mainly glaucoma and osteopenia. Pranayama, hatha yoga, and Bikram yoga were the most common yoga practices; headstand, shoulder stand, lotus position, and forceful breathing were the most common yoga postures and breathing techniques cited. Twenty-seven adverse events (35.5%) affected the musculoskeletal system; 14 (18.4%) the nervous system; and 9 (11.8%) the eyes. Fifteen cases (19.7%) reached full recovery; 9 cases (11.3%) partial recovery; 1 case (1.3%) no recovery; and 1 case (1.3%) died. As any other physical or mental practice, yoga should be practiced carefully under the guidance of a qualified instructor. Beginners should avoid extreme practices such as headstand, lotus position and forceful breathing. Individuals with medical preconditions should work with their physician and yoga teacher to appropriately adapt postures; patients with glaucoma should avoid inversions and patients with compromised bone should avoid forceful yoga practices.  相似文献

The synthesis of thromboplastin, a potent trigger of blood coagulation, can be induced in human peripheral blood monocytes. Indirect evidence suggests that newly synthesized thromboplastin becomes in part available on the cell surface. We have attempted to study the localization and availability of thromboplastin more directly by isolating plasma membranes from isolated human peripheral blood monocytes. The specific activities of the plasma membrane markers increased 16-22-fold in these preparations with a recovery of about 15%. The contamination by mitochondria, lysosomes, nuclei and endoplasmic reticulum was low as estimated by marker enzymes and electron microscopy. In both unstimulated and stimulated monocytes thromboplastin was largely recovered in this plasma membrane fraction, providing direct evidence for its membrane localization. Phospholipase C (E.C. 3.1.4.3) is a potent inactivator of thromboplastin through its hydrolysis of the phospholipids necessary for thromboplastin activity [Otnaess, Prydz, Bjørklid & Berre (1972) Eur. J. Biochem. 27, 238-243]. About 70% of the total membrane thromboplastin activity was inactivated when whole cells were treated with phospholipase C and the membranes subsequently isolated. Following stimulation to induce thromboplastin synthesis, the plasma membranes showed a shift in their relative content of phosphatidylcholine and phosphatidylethanolamine consistent with a transmethylation process.  相似文献

Aim

Climate change is expected to have major impacts on terrestrial biodiversity at all ecosystem levels, including reductions in species‐level distribution and abundance. We aim to test the extent to which land use management, such as setting‐aside forest from production, could reduce climate‐induced biodiversity impacts for specialist species over large geographical gradients.

Location

Sweden.

Methods

We applied ensembles of species distribution models based on citizen science data for six species of red‐listed old‐forest indicator fungi confined to spruce dead wood. We tested the effect on species habitat suitabilities of alternative climate change scenarios and varying amounts of forest set‐aside from production over the coming century.

Results

With 3.6% of forest area set‐aside from production and assuming no climate change, overall habitat suitabilities for all six species were projected to increase in response to maturing spruce in set‐aside forest. However, overall habitat suitabilities for all six species were projected to decline under climate change scenario RCP4.5 (intermediate–low emissions), with even greater declines projected under RCP 8.5 (high emissions). Increasing the amount of forest set‐aside to 16% resulted in significant increases in overall habitat suitability, with one species showing an increase. A further increase to 32% forest set‐aside resulted in considerably more positive trends, with three of six species increasing.

Main conclusions

There is interspecific variation in the importance of future macroclimate and resource availability on species occurrence. However, large‐scale conservation measures, such as increasing resource availability through setting aside forest from production, could reduce future negative effects from climate change, and early investment in conservation is likely to reduce the future negative impacts of climate change on specialist species.  相似文献
